My sister and I contribute towards my mom’s medical aid which is equivalent to R5 500 per month. This deduction comes out of my sister’s salary and as such, she is able to claim her medical contribution as a tax deduction.
In this case, will I be able to claim the portion that I contribute when I complete my tax return at the end of the year? Please advise.

As the contribution to your mother's medical aid is taken from your sister's account, she will be the one entitled to claim the medical aid tax credit.
The South African Revenue Service would allow your sister the credit, as they allow claims to be made in support of a dependent which does include parents. They do however look where the actual payment comes from.
What you could do to assist your situation is have your sister reimburse you half the monthly tax credit that she receives as a result of being allowed to claim this on her tax return.
